Buat suspensi kuman dengan NaCl fis ~stdr Mc
Farland 0,5.
Masukkan lidi kapas steril, oleskan pada
Muller Hinton Agar .
Letakkan Bacitrasin sebnyak 4 Buah.
Eramkan 24 jam 37°C.
Streptococcus pneumonia sensitive terhadap
optochin
Kelarutan
empedu:
1 cc Nacl+koloni sampai kekeruhan~Mc Farland.
+ 3-4 tetes larutan empedu(Na desoxycholate 10%)
Inkubasi 15 menit 37’C.
Hasil positifàlarutan jernih
Karena Pneumococcus berkapsul maka polisakarida dalam kapsul akan
larut dalam empedu.
Uji Inulin:
Biakan ditanam pada gula Inulin inkubasi 37’C 24 jam. Hasil positif pink->kuning.
